И еще, в каком буфере проводят раститровку антисывороток для Ухтерлони.
Может кто-то подскажет, реально работающую методику проведения иммунодиффузии для АФП.

Насколько я помню классикой является веронал-мединаловый. Однако народ последнее время меняет на фосфатный, чтобы с наркотиками и сильнодействующими не связываться. Даже на растворе Хэнкса делали. Однако АФП не делал. Может там какие нюансы.

1. Агар вернее агарозу в буфере просто так не растворить, надо плавить....
2. правильно написал Скабичевский тьфу Панаев классика "веронал-мединаловый" однако на деле можно пользовать и ПБС и трис-соль и физраствор.
3. Разницы в буферной системе конкретно для АФП ( альфа фетопротеина) нет. Пользовал ПБС.
4. Однако интересно почему детектируите АФП Ухтерлони, что получаите поликлоны ????
если хотите тестировать АФП в образцах надежнее делать ЕЛИСА. Если хотите оценить кол-во при выделении или вестерн или если есть тяга к классике рокетныий иммунофорез.

Агар плавится при низкой температуре в буфере (я пробовал трис-боратный) с 1М КI.
Совсем растворить не получается, если только в 1М щелочи, но сомневаюсь, что это вам поможет)
