Automatic recognition and isolation of single cells and cell colonies

The research on stem cells includes modifications on the genetic level in order to learn more about various regulations of genes responsible for self-renewal and the ability to differentiate into cells of all tissues. Recent findings indicate that stem cell colonies consist of mixed populations of cell types, with both transcriptional and temporal heterogeneity marking differentiation under all conditions. Analysis of expression profiles of stem cells on the single cell level allows for characterization of transcriptome dynamics and thereby provides a better understanding of stem cell differentiation. Single cell analysis can systematically determine unique cellular profiles for use in cell sorting and identification, show the potential to augment standing models of cellular differentiation, and elucidate the behaviour of stem cells exiting pluripotency.
Therefore, it is crucial to identify specific cells within the cell culture or sample using immunochemical staining or reporter gene systems in combination with targeted cell isolation. With the ability to harvest single cells in a reproducibly small amount of volume data from genetic analyis of single cells become more comparable. Recent technological advances have led to the development of automatic cell harvesting instruments like the CellCelector (AVISO). Automation and software controlled cell isolation not only facilitates the whole harvesting process but also allows for large-scale genetic analysis and expression profiling. As an alternative method to flow cytometry, which requires a large number of target cells, mechanically stresses cells and uses laser light for identification the CellCelector is a flexible tool for particularly gentle, individual cell isolation and exceptionally useful for single cell analysis.
