We have exciting projects on the potential of many human genes to be dual-coding (two overlapping open reading frames) or bicistronic (two non-overlapping open reading frames). Discover the true coding potential of our genome, study the structure/function of novel proteins, invent new tech. We use bioinformatics, proteomics, biochemistry and cell biology techniques. One (but not the only) of the novel proteins, alternative FUS, is co-expressed with the canonical FUS protein from the dual-coding FUS gene. altFUS coding sequence overlaps the FUS coding sequence in an alternative open reading frame. AltFUS is endogenously expressed in the human brain and other tissues, and both FUS and altFUS are involved in the aetiology of amyotrophic lateral sclerosis and its pathological hallmarks (PMID: 33226175). In this project, we are particularly interested in elucidating how altFUS prevents the recruitment of the PARKIN E3 ubiquitin ligase to the mitochondria during mitophagy. We also investigate large-scale protein-protein interactions and functionally annotate several thousands novel proteins in several species (PMID: 33179748).
